A Multiplex real-time PCR for detection of Mycoplasma gallisepticum and Mycoplasma synoviae in clinical samples from Brazilian commercial poultry flocks BJM

Mycoplasma gallisepticum (MS) and Mycoplasma synoviae (MS) are important avian pathogens and cause economic losses to the poultry industry. Molecular biology techniques are currently used for a rapid detection of these pathogens and the adoption of control measures of the diseases. The aim of this study was to develop and validate a technique for simultaneous detection of MG and MS by multiplex real time polymerase chain reaction (PCR). ABSTRACT The objective of this study was to identify the species and characterize the genetic relationships among mycoplasma isolates from commercial layer hen flocks using 16S-23S rDNA intergenic spacer region (IGSR) sequencing. Twenty-one isolates were obtained from samples collected from commercial layer flocks in four Brazilian states: São Paulo, Minas Gerais, Rio de Janeiro and Espírito Santo. The isolates were recovered from the São Paulo, Rio de Janeiro and Espírito Santo states. ABSTRACT This study aimed to compare method-based and newly developed sample-based methods for Mycoplasma gallisepticum (MG) detection in different samples of breeder flocks suffering from respiratory disease problems by using culture, real-time PCR (rPCR) and ELISA from chicks and embryonated eggs. Mycoplasma gallisepticum (Mg) infection of wild native Brazilian psittacines (Psittaciformes) which died of any cause during sorting, rehabilitation, or conservation, was investigated by PCR. Two previously described PCR methodologies using Mg specific primers were employed for the analyses of 140 swab samples (cloaca, trachea, or palatine cleft). Influence of culture medium components on Mycoplasma gallisepticum growth Rev Salud Anim.

The composition of a culture medium produced in Cuba to grow Mycoplasma gallisepticum was evaluated using a response surface methodology. Initially, the influence of Mycoplasma base, horse serum and yeast extract on biomass production was assessed in a 2³ factorial experimental design. Then, a central composite design was applied. Mycoplasma base and horse serum factors were selected for this study. In 7 L and 35 L bioreactors, the variations carried out in the culture medium composition were estimated. The experimental results were fitted to a second order polynomial model equation. Mycoplasmas, the simplest self-replicating organisms known, are distinguished phenotypically from other bacteria by their minute size and total lack of cell wall. The poultry industry is affected by several species of mycoplasmas, but Mycoplasma gallisepticum (MG) is the most economically significant one. The attachment of mycoplasmas to host respiratory epithelial cells constitutes a critical step in the pathway leading to infection and disease and is achieved by lipoproteins localized on the bacterial surface. Phenotypic and antigenic variation of Mycoplasma Gallisepticum vaccine strains BJM

Phenotypic and antigenic variations among Mycoplasma gallisepticum vaccine F and Ts-11 strains were investigated by SDS-PAGE and serological methods (hemagglutination-inhibition and immunoblot assays). The SDS-PAGE system followed by densitometer analysis showed weak phenotypic variability between the strains, being the major difference close to the 75 kDa level where a prominent peptide band was detected only in the F vaccine strain. Polyclonal antibodies to the M. gallisepticum antigens produced in chickens were used in serological tests for this antigenic variability study. The Mycoplasma gallisepticum strains [wild-type S6 (208) and a vaccine-type F-K810] grown in Frey´s and Hayflick´s media were analyzed by SDS-PAGE. No visual changes in the protein profiles of these strains were observed regardless of the media composition used, although the polyacrylamide gel electrophoretograms showed minor differences do exist when densitometer traces of the gel are compared. Both strains were easily differentiated on SDS-PAGE analysis by a peptide band p75, that is specific for MG F-K810 strain, used as vaccine.

Avian salmonellosis and mycoplasmosis are infectious diseases that, in addition of causing lack of flock uniformity, represent a hazard to human health. The objective of the present study was to evaluate the seroprevalence of mycoplasmosis and salmonellosis in commercial broilers, backyard chickens, and spent hens slaughtered at a processing plant with local health inspection in Uberlândia, MG, Brazil.
